The GS piggyBac® transposon system

The GS piggyBac® system uses an engineered hyperactive piggyBac® transposase enzyme to insert expression vector cargos with your gene of interest into the host cell genome with high efficiency, supporting stable piggyBac® integration, with the following benefits:

  • Enhanced performance of the GS piggyBac® system is observed with low versus high expressing monoclonal antibodies
  • Accommodating a large DNA cargo (> 200kb) makes the GS piggyBac® system highly suited to support cell line engineering needed to enhance complex protein expression
  • Preferentially targeting genetically stable parts of the genome at high efficiency has the potential to improve cell line stability

The GS piggyBac® transposon system is Lonza’s advanced cell line engineering technology that uses a hyperactive piggyBac® transposase to integrate expression vector cargos into stable regions of the genome. This supports high efficiency piggyBac® integration, robust gene expression, and scalable bioproduction processes.

The piggyBac® system enables efficient cell line engineering by preferentially inserting genetic material into stable, transcriptionally active genomic regions. This improves expression stability, supports large DNA cargos, and accelerates development timelines when it is used in conjunction with the GS Gene Expression System® technology platform.

Unlike random or low efficiency integration methods, the piggyBac® transposon system targets genetically stable genomic regions associated with high expression.

  • More predictable cell line performance
  • Higher titers
  • Greater stability across scaleup
  • Reduced development risk

Absolutely. The piggyBac® system can integrate DNA cargos exceeding 200 kb, making it ideal for engineering cell lines that express complex proteins, multigene pathways, or advanced therapeutic formats.
