Abstract:
The production of New Molecular Format (NMF) proteins requires innovative methods to increase the productivity of the bioreactor process while ensuring consistent product quality. In this study, the effects of Lonza's high inoculation density (HID) fed-batch process on the productivity and product quality characteristics of two CHO cell lines expressing unique NMFs, a monospecific antibody with an Fc fusion protein and a bispecific antibody, were investigated in comparison to low inoculation density (LID) fed-batch processes. It was found that the intensified platform fed-batch process increased product concentrations for the two uniquely structured complex proteins by 33 and 109%, respectively, at shorter culture times, while maintaining similar product quality characteristics to conventional fed-batch processes.
