Lentiviral vectors (LVVs) are increasingly needed for gene and cell therapies, but transient transfection limits scalability and consistency. To address this need, we developed an inducible stable LVV producer cell line using a CD19 CAR as the gene‑of‑interest. From the monoclonal research cell banks generated by our workflow, the top clones reached up to 5 × 10⁷ TU/mL and maintained stable productivity for more than 20 passages, supporting scale‑up beyond 2000 L. The resulting PCL‑derived LVVs also demonstrated higher infectivity and potency compared with transient‑transfection material. These results confirm a high‑yield, GMP‑aligned LVV PCL platform suitable for generating robust, scalable lines for CAR and other therapeutic genes.
